Summary :
Quantitative EEG analyses of long-term derivations (4 hour-period, starting 30 minutes after drug intake) clearly demonstrated a dose related pharmacodynamic effect and consequently the bioavailability of EGb 761.
Compared to placebo the following pharmaco-dynamic changes observed in the EEG derivations commenced during the first hour after drug intake and lasted at lease 4.5 hours : stabilisation of vigilance with 120 mg ; quasi-uniform vigilance with 240 mg ; a hyper-vigilance state with 600 mg, commencing 110 minutes after drug intake ; 400 mg gave intermediate results confirming these pharmacodynamic effects ; a dose-related progressive increase in the power of the dominant frequency, with an increase in the absolute value of alpha and beta-1 dominant frequencies in the frontal derivations.
