This method has been applied to the evaluation of both narcotic (meperidine, morphine, levorphan and nalorphine) and non-narcotic (aspirin, sodium salicylate, paracetamol aminopyrine, phenylbutazone and oxyphenbutazone) analgesics.
It separates the two groups by bringing out the greater potency and shorter latency of the former.
Aminopyrine and nalorphine exert no analgesic action against bradykinin-evoked pain at doses of 46 % of the LD50.
Among the non-narcotic analgesics, aspirin proved to be the safest if not the most effective from the standpoint of relative toxicity, the 50% Acute Anti-Chemoalgesic Dose (A-ACAD50, which refers to the chemical nature of the bradykinin stimulus) being 7.3% of the LD50.
This compares with paracetamol 13%, phenylbutazone and oxyphenbutazone 6%, and sodium salicylate 35% LD50.
On the other hand, the A-ACAD50 of the narcotics expressed in terms of toxicity, morphine 0.3%, levorphan 1.4% and meperidine 1.8% LD50, are all considerably smaller than that of aspirin.
This difference between the two groups of analgesics is reflected in the low analgesic dose of the narcotic analgesics on a weight basis.
